1,2,3-Triazoles Fused to Aromatic Rings

The structure, synthesis, reactivity and applications of 1,2,3-triazoles fused to aromatic rings are described. These compounds have been classified in two groups by a structural approach: (a) fused 1,2,3-triazoles without a bridgehead nitrogen atom and (b) fused 1,2,3-triazoles with a bridgehead nitrogen atom. Although both systems present a similar structure, the synthetic procedures and their reactivity are different.

The crystal structure of 1 consists of one [Cu(tn)]2+ unit and one tcpd2- anion, both located on general positions. The first isolation of an antimony(III) thiocyanate complex salt, [(CH3)4N][Sb(CNS)4] (I), and of two adducts of the Sb(CNS)3 formal unit with potentially tridentate nitrogen donors, Sb(CNS)3tpta (II) and [Sb(CNS)3]3(dqp)2 (III), is described. In these derivatives, infrared and Mossbauer spectral data are essentially indicating the predominant presence of N-bonded thiocyanate. For complex I, the occurrence also of groups bridging through the nitrogen atom is advanced. Polyaza[n]paracyclophanes as synthetic models of Zn containing enzymes. The role of a non coordinated nitrogen atom in the proximity of the metal

Abstract 2,6,9,13-Tetraaza[14]paracyclophane ( 3, B323 ) represents a simple model for Zn containing enzymes like HCAII which possess additional reactive groups in the proximity of the metal center. The Zn 2+ . 3 complex shows notable catalytic activity for the hydrolysis of p -nitrophenyl acetate. Structural modifications of 3 which affect to the non coordinated nitrogen atom greatly modify the catalytic activity of the receptor.

Reactions of Piperazin-2-one, Morpholin-3-one, and Thiomorpholin-3-one with Triethyl Phosphite Prompted by Phosphoryl Chloride: Scope and Limitations

The reaction of the title lactams with triethyl phosphite prompted by phosphoryl chloride provided six-membered ring heterocyclic phosphonates or bisphosphonates. These novel scaffolds might be of interest as building blocks in medicinal chemistry. The course of the reaction was dependent on the structure of the used substrate. Thus, morpholin-3-one and thiomorpholin-3-one readily provided the corresponding 1,1-bisphosphonates (compounds 1, 2, 7, 14 and 16), whereas the protection of their nitrogen atom resulted in the formation of dehydrophosphonates (compounds 5, 6, and 8). Base-Dependent Stereodivergent Intramolecular Aza-Michael Reaction: Asymmetric Synthesis of 1,3-Disubstituted Isoindolines

The nucleophilic addition (A(N)) / intramolecular aza-Michael reaction (IMAMR) process on Ellman's tert-butylsulfinyl imines, bearing a Michael acceptor in the ortho position, is studied. This reaction affords 1,3-disubstituted isoindolines with a wide range of substituents in good yields and diastereoselectivities. Interestingly, careful choice of the base for the aza-Michael step allows either the cis or the trans diastereoisomers to be exclusively obtained. This stereodivergent cyclization has enabled the synthesis of C2-symmetric bisacetate-substituted isoindolines. An efficient entry to optically active anti- and syn-beta-amino-alpha-trifluoromethyl alcohols.

The reaction of chiral 5,6-dihydro-2H-1,4-oxazin-2-ones with TMSCF3 in the presence of a suitable activator leads to trifluoromethyl lactols, which can be selectively reduced to anti-beta-amino-alpha-trifluoromethyl alcohols. The corresponding syn diastereoisomers are obtained when the starting imines are reduced and the nitrogen atom is conveniently protected. In addition, a novel rearrangement of the CF3 group in the lactol intermediates has been observed. This represents a formal CF3 addition to the imine function in the starting substrates.

Convergent results from experimental and theoretical DFT studies of the intramolecular rearrangement of Z-hydrazones of 3-acetyl-1,2,4-oxadiazoles

A combined kinetic and theoretical study of the monocyclic rearrangements of heterocycles (MRH) has been carried out. The interconversion of the Z-hydrazone of 3-benzoyl-5-phenyl-1,2,4-oxadiazole into the corresponding triazole has been experimentally investigated in dioxane/water in the pS(+) range 5.5(5)-13.9. The uncatalyzed region has been examined at the DFT level using a model system formed by the Z-hydrazone of 3-formyl-1,2,4-oxadiazole and one or two water molecules. The environmental effect of the solvent has been emulated using a continuum model (COSMO) approach.
